Semax is a synthetic heptapeptide derived from the 4–10 fragment of adrenocorticotropic hormone (ACTH), modified with a C-terminal Pro-Gly-Pro sequence intended to improve its stability. In the literature it is grouped with the neuropeptide research compounds.

How is Semax’s mechanism studied in research?

Semax has been examined in neurobiological research, primarily in animal models:

  • Neurotrophin signalling — studied in relation to brain-derived neurotrophic factor (BDNF) and its receptor TrkB.
  • Neuropeptide activity — investigated as a stabilised ACTH(4–10) fragment.

The animal-model literature is still active: a 2025 study in Acta Naturae examined Semax and a Semax derivative in a transgenic Alzheimer’s-model mouse line, and a 2025 cellular study in the Bulletin of Experimental Biology and Medicine investigated its action on intracellular calcium dynamics in rat neurons. What compounds is Semax studied alongside?

In the published literature, Semax is studied within the same regulatory-peptide research programme as Selank, with one functional-connectivity study examining the two side by side, and a 2024 animal-model study tested it together with melanotan II as a fellow synthetic ACTH(4–10) analogue.
