Cagrilintide is a synthetic amylin analogue carrying amino-acid substitutions that improve solubility and stability, plus a lipidation that extends its circulating half-life through albumin binding. Its development as a long-acting analogue was described by Kruse and colleagues in Journal of Medicinal Chemistry in 2021.

How is Cagrilintide’s mechanism studied in research?

Amylin acts at receptor complexes formed by the calcitonin receptor together with receptor-activity-modifying proteins (RAMPs) — the AMY receptors.
